#141: Versatile T7 RNA Polymerases that Generate Less Immunostimulatory mRNA

Our poster explores the use of T7 RNA polymerase (T7 RNAP) in in vitro transcription (IVT) for mRNA synthesis—a well-established method for producing high quantities of synthetic RNA. Despite its efficiency, T7 RNAP can produce immunostimulatory byproducts like double-stranded RNA (dsRNA). Addressing these challenges, we've developed a series of engineered T7 RNAP mutants. Our findings highlight their enhanced processivity and yield, alongside a significant reduction in undesirable byproducts compared to both wild-type and other commercially available enzymes.
